Use toyota genuine manual transmission fluid then check. Else live with it, old gear box usually whine.
Completely flush the gear oil and add sae 80 or 75w-85 gl4 spec oil ....old gearbox usually whine a bitBut notice at which gear whine occurs.......if whine occurs only when clutch is released then its input shaft bearing if it's in all gears except 4th gear then counter shaft bearing........if comes at very high speeds then differential gear is worn
Mine also do the same only going uphill. When gearbox is under load. Otherwise it does not whine.
check and replace the gear oil first! most probably the noise will reduce after the oil change
old school method, just add one banana into the oil intake plug, usually it is at passenger side wheel, right behind the water splash shield.

How have you localized the noise to the gearbox?
It comes in 5th gear usually upon accelerating it makes loud whine noise even after inspection mechanic said its from gearbox, in neutral.giving revs nothing comes out.so.its gearbox
Is it just the 5th gear? Are the other gears quiet? Is it only on acceleration? It could be a bearing on just the differential backlash out of specification.
I agree that this is the correct approach to troubleshoot the problem.
It could be one of the gearbox bearings starting to get noisy.
So should i go for a bearing replacement by opening it? New bearings are available?

Changing gearbox or getting in overhaul which is very hard is the only solution
