Probably not since big chunks of road are inundated. Also, with the current unfortunate circumstances prevailing in Sindh, it's not safe.

Karachi to Hyderabad via Motorway
Then Hyderabad to Sukkur via N5
Then Sukkur to Multan Motorway
All clear

2 days back one of my colleague travelled from Bahawalpur to Karachi and this is what he said:

People who are planning to travel to Karachi or Sindh by road, please avoid. Worst conditions.
It took me 19 hours from Bahawalpur to reach only Moro.

Bahawalpur to Sukkar - 6 hours (M5 motorway)
Sukkur to Moro - 13 hours (160km)
