Lahore to Karachi via road

I am planning to travel to karachi from Lahore via road.

I am a single driver and plan to do it in one go.

What route should i take?
where to stop for refuelling etc
Is it safe to travel Sukkhur to Karachi in evening night? or we should do it in day light?

If you want to do it in one go and its your first time driving by road upnorth, then i will suggest you to leave karachi by 4am so that you reach hyderabad easily by 6am and it would be daylight so it would be easier to drive on the national highway.
From hyderabad onwards there are two routes, one is the normal national highway and the other is known as the sehwan wala route. It is a single road but is recently built so the route is good and it also saves 20/30 minutes if you travel by national highway. But i wont recommend you to use sehwan route in the night because it gets extremely dangerous as many drivers overtake illegaly..
For refueling you can use any pump, fill your tank up from karachi and it should easily go till sukkur, before sukkur there is a shell pump which is recently built with clean washrooms etc so wont be a problem. Before getting on sukkur-multan motorway you should fill up your tank which should easily take you to multan. From multan you can exit the motorway and refuel again although from multan onwards rest areas are operational. You should take stops after every 150kms and listen to songs etc or you should have someone to talk to so that you dont fall asleep.
Rest details you can easily get from youtube as many people have done khi-isl by road and uploaded videos with complete details

Now adays it is very much safe to travel during the night. not an issue!

Honda civic (fuel tank capacity = 52L, if i am not wrong) means you can easily cover 500 kms in a single run.

Route:
Lahore to Multan through M3 & M4 (approx 330 kms)
Multan to Sukkur through M5 (approx 410 kms)
Sukkur to Hyderabad through N5 (approx 320 kms)
Hyderabad to Karachi through M9 (approx 160 kms)

Guide:
I suggest you to take 2 stops. Leave Lahore at 4 AM, you can easily reach Multan around 8 AM, leave Multan at 9 AM and reach Sukkur around 1 PM, leave Sukkur at 1:30 PM and reach Karachi around 8 PM. At both stops, you can take a rest of 30 minutes, refresh yourself and other passengers, refuel your tank. i am attaching Google Maps Links of both the locations for your help.

at both places you can easily find fuel, food and neat washrooms.

It is safe to travel between Sukkur and Karachi in night but i still suggest you to travel this portion in daylight.
