Can some one tell if the newly built part of M4 (toba) is open for traffic? What shall be the best point to connect M4 while traveling from Multan to ISLAMABAD?

Toba to Shorkot is not operational yet...
you can select M4 portion from Multan to Khanewal then came through Kabirwala, Shorkot, Jhang and then touch M4 again from Gojra. Road from Khanewal till Gojra is 6/10, Single track with pot holes..
Then from Gojra you can drive over M4/M2 till Islamabad.

In my opinion, Khanewal-Chichawatni-Kamali-Toba-Gojra-M4 is better route than Khanewal-kabirwala-shorkot-jhang-M4

as discussed in earlier, khanewal-chichawatni-kamalia in the best route. If you want to go from motorway then from kamalia-toba-gojra and m4 to faisallabad.

Thank you for your suggestion I like to prefer Khanewal-Mian Chunno-Chichawatni-Kamalia to Faisalabad route,
one thing more is it safe to drive in the night after 8 pm with family onboard and what about the road conditions in this route as I am traveling from Karachi

Security situation is same as other areas of Central Punjab. Take routine safety precautions as advisable in Pakistan. It is also advisable not to travel on deserted roads especially at dawn or dusk, and overnight if possible. Also during dark hours avoid travelling on bypasses. Toba Tek Singh has a bypass which should be preferred during day time.
