Its basically Indus Highway (N-55), which, North Bound, practically starts from Jamshoro goes to PWR via DADU-LARKANA-KASHMORE-DG KHAN-DI KHAN-KOHAT-PWR and shortest way to reach PWR or Islamabad for that matter. (I reached Bannu in 15.5 hrs, 1380 KM but that was crazy since drove all the way alone n virtually non-stop), route was Karachi-Ubauro (N-5) and then Guddu Barrage-Kashmore then N-55.
For D.I. Khan to Chashma, its not highway but a single road (one up/one down way only, no hard shoulder/overtaking lane etc). Either you can take Chashma link road right in the middle of DI Khan (just ask anybody) or you can go past DI KHAN (12-15 KM) and then there is a link road on your right going towards Paniala (Maulana Fazl-ur-Rehman aka Maulana Diesel hometown) towards Chashma. and then cross Chanshma barrage bridge, town, Nuclear plant and join M'garh/Mianwali road at Chandni Chowk), turn left for ISB and right for KHI.
Nowadays due to Taliban trouble, its not recommended and do not try to attempt this (DI KHAN-CHASHMA Section) in the night otherwise you will get lost and there are no signs or people because population is scarce to help you.
